Study Shows Aerobic Exercise Helps Cognitive Function in Older Adults A study in Frontiers in Endocrinology explored how exercise could improve cognitive function in older adults, reports Florida Atlantic University (FAU). The researchers considered three specific biomarkers expected to increase in older adults following exercise training and correlate with cognition and metabolomics markers of brain health: myokine Cathepsin B (CTSB), brain derived neurotrophic factor (BDNF)..., and klotho, in addition to metabolomics, which have become increasingly used to understand biochemical pathways that may be impacted by Alzheimer's disease (AD). A metabolomics analysis was performed on blood samples of 23 asymptomatic late middle-aged adults with familial and genetic risk for AD. Participants were split into cohorts assigned to perform usual physical activity and enhanced physical activity (EPA), with the latter undergoing 26 weeks of supervised treadmill training. After 26 weeks, plasma CTSB levels were higher in adults at risk for AD in the EPA group. Verbal learning and memory also had a positive correlation with change in CTSB but was not related to BDNF or klotho. Meanwhile, plasma BDNF levels declined in conjunction with metabolomic changes, including reductions in ceramides, sphingo- and phospholipids, and changes in gut microbiome metabolites and redox homeostasis. "The positive association between CTSB and cognition, and the substantial modulation of lipid metabolites implicated in dementia, support the beneficial effects of exercise training on brain function and brain health in asymptomatic individuals at risk for Alzheimer's disease," concluded FAU's Henriette van Praag.
